Case Isescalation revert back to False.

Case escalation checkbox revert back to false when user is changed.can anyone help me out why this things happened.In my org. no escalation rule defined.

This is the default behaviour.
However, you can have a workflow or trigger which changes the IsEscalated field back to falsebased on some condition.
