Date/Time Field to Calculate in Seconds

Help!!! Here is my scenario - I need to calculate in SECONDS a duration of time as follows;
 
Start Time:  2:01:25
End Time: 2:02:10
Duratation Start Time - End Time with Seconds
 
Can this be done and has anybody ever done it?
