 I don't think you would find one comprehensive guide, These are serveral topics in service cloud

if you are talking to start with you need service cloud licences if you want to try in developer edition go to the logged in use details and select the check box Service Cloud User then you will get an option under customize saying that Service Cloud Console...then start setting up that and start using the concepts like Case Management, Live agent, CTI, Contact Center setup, Entitlements , Answers, Social Media integration.....etc.... features are here http://www.salesforce.com/service-cloud/features/
