While it's not clear what kind of testing you wish to do, you can start with exploring the SOAP API with the online documentation found here: http://www.salesforce.com/us/developer/docs/api/index.htm, or if you need a REST approach, there's similar documentation found here: http://www.salesforce.com/us/developer/docs/api_rest/index.htm

There is a substantial amount of content regarding connecting to and using the various API's on developer.force.com, but you'll have to search for it based on what you want to accomplish.

What kind of org are you connecting to and what is the user license of the user whose credentials you are using? Do they have a profile where the API Enabled permission is checked?
